腫瘤代謝及細(xì)胞極性的分子調(diào)控機(jī)制
1. 深入探討上皮細(xì)胞中的極性基因功能以及在上皮屏障中的重要分子機(jī)制,上皮細(xì)胞命運(yùn)調(diào)控過程中極性信號的關(guān)鍵機(jī)制,比如在自噬以及壞死性凋亡(necroptosis)中的調(diào)控機(jī)制,并探討上皮細(xì)胞屏障破壞和和修復(fù)過程中極性基因的調(diào)控機(jī)制,以及上皮細(xì)胞屏障破壞與腫瘤發(fā)生的作用機(jī)制研究。
2. 以腫瘤原位移植技術(shù)結(jié)合生物信息學(xué),分子生物學(xué)方法,獲得腫瘤在肺部轉(zhuǎn)移和定值中的關(guān)鍵分子節(jié)點(diǎn),上皮細(xì)胞EMT和MET轉(zhuǎn)化過程中極性信號的調(diào)控機(jī)制,以及在遠(yuǎn)端轉(zhuǎn)移中的相關(guān)機(jī)制。
3. 以目前先進(jìn)的類器官技術(shù)(organoid),進(jìn)一步探討上皮細(xì)胞穩(wěn)態(tài)(Homeostasis)打破與損傷修復(fù)中極性通路的新功能,來研究腫瘤早期發(fā)生中與腫瘤微環(huán)境的相互作用,從而明確極性信號在決定上皮細(xì)胞命運(yùn)以及異質(zhì)性中的分子機(jī)制。
