腫瘤細胞可塑性的表觀遺傳調(diào)控

研究內(nèi)容

可塑性調(diào)控在藥物抵抗和腫瘤轉(zhuǎn)移中發(fā)揮著至關(guān)重要的作用,同時也是導(dǎo)致腫瘤異質(zhì)性的主要原因之一??伤苄哉{(diào)控可被理解為細胞改變其身份的過程,包括同一譜系下細胞的分化,或者是細胞譜系的轉(zhuǎn)分化或去分化(例如,針對肺癌和前列腺癌的靶向治療導(dǎo)致下腺癌表型的轉(zhuǎn)變?yōu)樯窠?jīng)內(nèi)分泌腫瘤,以及干細胞的惡性轉(zhuǎn)化、腫瘤EMT和MET在腫瘤轉(zhuǎn)移過程中的動態(tài)變化等)。進一步可塑性調(diào)控也可擴展至腫瘤整體表型的轉(zhuǎn)變,例如惰性腫瘤的進展演化以及轉(zhuǎn)移過程中腫瘤的休眠和蘇醒等現(xiàn)象。
我們的研究以表觀遺傳調(diào)控與腫瘤細胞命運決定及微環(huán)境重塑為核心,聚焦前列腺癌及腸癌診治中的臨床重大需求,探究表觀遺傳調(diào)控在腫瘤轉(zhuǎn)移及耐藥中的功能,并基于此提出潛在的治療策略;闡明表觀遺傳相關(guān)基因如何響應(yīng)藥物治療、代謝、炎癥等信號調(diào)控組蛋白修飾及染色質(zhì)重塑進而決定腫瘤及微環(huán)境細胞的命運及功能。
